Four complexes of the type [Mn(dpa)2(X)2] ·H2O (dpa = 2,2?-dipyridylamine; X = N3 -,1; X = NCO-, 2) and [Mn(dpa)2(X)(OH 2)]ClO4 (X = N3-, 3; X = NCO -, 4) have been synthesized using single-pot reactions of manganese(II) salt, dpa and X in 1:2:2 and 1:2:1 molar ratios, respectively. The compounds have been characterized by microanalytical, physicochemical, spectroscopic and magnetic results. Room temperature magnetic moments (mueff) are in the range 5.88-5.97 BM and are typical of high-spin manganese(II) complexes (t2g5 idealized, S = 5/2). The complexes display a Mn(III)-Mn(II) couple at ?0.6 V versus SCE in DMF solutions. X-ray structure determinations show that in 1-4 the manganese(II) centres are in a distorted octahedral environment. Compounds 1 and 2 contain the MnN6 chromophore, completed by two bidentate dpa units and two terminal azido groups in 1 or N-coordinated cyanato ligands in 2, along with a water of crystallization. In 3 and 4, one water molecule acts as a coordinating ligand replacing one of the terminal pseudohalides in 1 and 2, respectively, resulting in a MnN5O chromophore. The amine H, terminal azido N or cyanato O, O and H’s of the water molecule are engaged in extensive hydrogen bonding of the type N-H?N, O-H?N and N-H?O in 1 and 2. Additionally O-H?O bonding is observed in 3 and 4. Supramolecular 2D sheet structures are obtained arising from such weak intermolecular forces. All the complexes display high-energy intraligand 1(pi-pi*) fluroscence and intraligand 3(pi-pi*) phosphorescence in glassy solutions (DMF at 77 K).

Glycosynthase Principle Transformed into Biocatalytic Process Technology: Lacto-N-triose II Production with Engineered exo-Hexosaminidase

Glycosynthases are promising enzyme catalysts for glycoside synthesis. Derived from glycoside hydrolases by mechanistic repurposing of their active site, glycosynthases utilize suitably activated glycosyl donors for glycosylation, yet they are unable to hydrolyze the products thus formed. Although primed for synthetic application by their design, glycosynthases have yet to see actual use in carbohydrate production. To challenge limitations on glycosynthase applicability perceived from the process chemistry point of view, here we developed a glycosynthase (D746E variant) from Bifidobacterium bifidum beta-N-acetylhexosaminidase that is highly active synthetically (?100 mumol min-1 mg-1) and fully chemo- and regioselective when using N-acetyl-d-glucosamine 1,2-oxazoline for beta-1,3-glycosylation of lactose. We thus established a chemoenzymatic process technology for production of lacto-N-triose II, a core structural unit of human milk oligosaccharides. Using equivalent amounts of oxazoline (prepared chemically in 40% yield from N-acetyl-d-glucosamine) and lactose, we obtained lacto-N-triose II (515 mM; 281 mg mL-1 90% yield; ?1 h reaction time) immediately recoverable from the reaction in 85% purity. These metrics of process efficiency reveal the prodigious potential of the glycosynthase for trisaccharide production.

Low-budget 3D-printed equipment for continuous flow reactions

This article describes the development and manufacturing of lab equipment, which is needed for the use in flow chemistry. We developed a rack of four syringe pumps controlled by one Arduino computer, which can be manufactured with a commonly available 3D printer and readily available parts. Also, we printed various flow reactor cells, which are fully customizable for each individual reaction. With this equipment we performed some multistep glycosylation reactions, where multiple 3D-printed flow reactors were used in series.

Pocket-based Lead Optimization Strategy for the Design and Synthesis of Chitinase Inhibitors

Insect chitinases play an indispensable role in shedding old cuticle during molting. Targeting chitinase inhibition is a promising pest control strategy. Of ChtI, a chitinase from the destructive insect pest Ostrinia furnacalis (Asian corn borer), has been suggested as a potential target for designing green pesticides. A 4,5,6,7-tetrahydrobenzo[b]thiophene-3-carboxylate scaffold was previously obtained, and further derivatization generated the lead compound 1 as Of ChtI inhibitor. Here, based on the predicted binding mode of compound 1, the pocket-based lead optimization strategy was applied. A series of analogues was synthesized, and their inhibitory activities against Of ChtI were evaluated. Compound 8 with 6-tert-pentyl showed preferential inhibitory activity with a Ki value of 0.71 muM. Their structure-activity relationships suggested that the compound with larger steric hindrance at the 6-nonpolar group was essential for inhibitory activity due to its stronger interactions with surrounding amino acids. This work provides a strategy for designing potential chitinase inhibitors.

A simple, mild, and cost effective methodology has been developed for the synthesis of aryl thio-and selenoglycosides from glycosyl halides and diaryl dichalcogenides. Diaryl dichalcogenides undergo reductive cleavage in the presence of rongalite (HOCH2SO2Na) to generate a chalcogenide anion in situ followed by reaction with glycosyl halides to furnish the corresponding aryl thio- and selenoglycosides in excellent yields. Using this protocol, synthesis of 4-methyl-7-thioumbelliferyl-beta-d-cellobioside (MUS-CB), a fluorescent non-hydrolyzable substrate analogue for cellulases has been achieved.

Dichloro-cyanoacetimidates as glycosyl donors

Transformation of 1-O-unprotected glucose and galactose derivatives (1a-d) into O-glycosyl dichloro-cyanoacetimidates (2a-d) was performed with dichloro-cyanoacetonitrile in the presence of DBU as base. Reaction with different acceptors (3a-d) under TMSOTf catalysis afforded glycosides 4 in high yields. Competition experiments with O-glucopyranosyl trichloroacetimidate 10a, bearing a 4-tert-butylbenzyl group at 6-O, and O-glucopyranosyl dichloro-cyanoacetimidate 10b, bearing a 4-methylbenzyl group at 6-O, displayed similar reactivities for these two types of glycosyl donors.

Luminescent two-dimensional coordination polymer for selective and recyclable sensing of nitroaromatic compounds with high sensitivity in water

A stable two-dimensional (2D) coordination Cd(II) polymer [Cd(ppvppa)(1,4-NDC)]n (1) (ppvppa = N-(pyridin-2-yl)-N-(4-(2-(pyridin-4-yl)vinyl)phenyl)pyridin-2-amine; 1,4-H2NDC = 1,4-naphthalenedicarboxylic acid) was prepared by the solvothermal reactions of Cd(NO3)2·4H2O with ppvppa and 1,4-H2NDC in H2O and MeCN at 150 C. Compound 1 was characterized by elemental analysis, IR, powder X-ray diffraction (PXRD), and single crystal X-ray diffraction and thermogravimetric analysis (TGA). Compound 1 consists of dimeric [Cd2(ppvppa)2] fragments linked by 1,4-NDC bridges to generate a 2D honeycomb network. Complex 1 strongly emits blue light in the solid state when excited at 419 nm at ambient temperature. This emission is selectively quenched by nitro aromatic compounds in water with good sensitivity. Compound 1 also displays recyclable detection of nitro aromatic compounds.

We report the IR and UV-vis spectra of three bridgehead silanimines, 1-sila-2-azabicyclo[3.2.2]non-1(2)-ene, 1-sila-2-azabicyclo[3.2.1]oct-1(2)-ene, and 1-sila-2-azabicyclo[2.2.2]oct-1(2)-ene, prepared in matrix isolation by UV irradiation of bridgehead silyl azide precursors and characterized by chemical trapping with trimethoxymethylsilane with and without prior selective photodestruction with light of suitable wavelengths. The IR spectra of the 14N and 15N species and the n ? pi* transition in their UV-vis spectra are in good agreement with ab initio computations. It is concluded that pi bonding plays a crucial role in determining the properties of silanimines.

Statin Lactonization by Uridine 5?-Diphospho-glucuronosyltransferases (UGTs)

Statins are cholesterol-lowering drugs that have proven to be effective in lowering the risk of major cardiovascular events. Although well tolerated, statin-induced myopathies are the most common side effects. Compared to their pharmacologically active acid form, statin lactones are more potent inducers of toxicity. They can be formed by glucuronidation mediated by uridine 5?-diphospho-glucuronosyltransferases (UGTs), but a systematic characterization of subtype specificity and kinetics of lactonization is lacking. Here, we demonstrate for six clinically relevant statins that only UGT1A1, 1A3, and 2B7 contribute significantly to their lactonization. UGT1A3 appeared to have the highest lactonization capacity with marked differences in statin conversion rates: pitavastatin atorvastatin > cerivastatin > lovastatin > rosuvastatin (simvastatin not converted). Using in silico modeling we could identify a probable statin interaction region in the UGT binding pocket. Polymorphisms in these regions of UGT1A1, 1A3, and 2B7 may be a contributing factor in statin-induced myopathies, which could be used in personalization of statin therapy with improved safety.

Synthesis, structural, thermal characterization and interaction with calf-thymus DNA and albumins of cationic Ni(II) complexes with 2,2?-dipyridylamine and salicylaldehydes

Five cationic mixed-ligand Ni(II) complexes with 2,2?-dipyridylamine (dpamH) and substituted salicylaldehydes (X-saloH), having the general formula [Ni(dpamH)2(X-salo)](NO3), were prepared and characterized by physicochemical and spectroscopic techniques (FT-IR and UV?vis) and their interaction with calf-thymus (CT) DNA and serum albumins was investigated. The complexes are formulated as [Ni(dpamH)2(3-OCH3-salo)](NO3)·CH3OH, 2, [Ni(dpamH)2(5-CH3-salo)](NO3)·CH3OH, 3, [Ni(dpamH)2(5-NO2-salo)](NO3)·CH3OH, 4, [Ni(dpamH)2(5-Cl-salo)](NO3), 5 and [Ni(dpamH)2(5-Br-salo)](NO3), 6. Moreover, in the absence of dpamH, one neutral nickel-salicylaldehydato complex was also prepared and characterized as [Ni(5-NO2-salo)2(CH3OH)2], 1. The structures of complexes 2 and 4 were verified by X-ray crystallography. The thermal stability of the complexes was examined by the simultaneous thermal analysis technique (TG/DTG-DTA). Spectroscopic (UV?vis), electrochemical (cyclic voltammetry) and physicochemical (viscosity measurements) techniques were employed in order to study the binding mode and binding strength of the complexes to CT DNA, while ethidium bromide (EB) displacement studies (performed by fluorescence emission spectroscopy) revealed the ability of the complexes to displace the DNA-bound EB. Intercalation is the most possible mode of interaction of the complexes with CT DNA. The interaction of the complexes with albumins was studied by fluorescence emission spectroscopy and the determined binding constants present relative high values.
