N-acetylglucosamine, the building block of chitin, inhibits growth of Neurospora crassa

N-acetylglucosamine (GlcNAc) is the monomer of the polysaccharide chitin, an essential structural component of the fungal cell wall and the arthropod exoskeleton. We recently showed that the genes encoding the enzymes for GlcNAc catabolism are clustered in several ascomycetes. In the present study we tested these fungi for growth on GlcNAc and chitin. All fungi, containing the GlcNAc gene cluster, could grow on GlcNAc with the exception of four independent Neurospora crassa wild-type isolates, which were however able to grow on chitin. GlcNAc even inhibited their growth in the presence of other carbon sources. Genes involved in GlcNAc catabolism were strongly upregulated in the presence of GlcNAc, but during growth on chitin their expression was not increased. Deletion of hxk-3 (encoding the first catabolic enzyme, GlcNAc-hexokinase) and ngt-1 (encoding the GlcNAc transporter) improved growth of N. crassa on GlcNAc in the presence of glycerol. A crucial step in GlcNAc catabolism is enzymatic conversion from glucosamine-6-phosphate to fructose-6-phosphate, catalyzed by the glucosamine-6-phosphate deaminase, DAM-1. To assess, if DAM-1 is compromised in N. crassa, the orthologue from Trichoderma reesei, Trdam1, was expressed in N. crassa. Trdam1 expression partially alleviated the negative effects of GlcNAc in the presence of a second carbon source, but did not fully restore growth on GlcNAc. Our results indicate that the GlcNAc-catabolism pathway is bypassed during growth of N. crassa on chitin by use of an alternative pathway, emphasizing the different strategies that have evolved in the fungal kingdom for chitin utilization.

The title compound, (Hdipya)(H3pyr), where Hdipya is protonated 2,2′-dipyridilamine and H3pyr is the monoanion of pyromellitic acid (H4pyr), was obtained from a reaction mixture containing Zn(II) ions, dipya, Na4pyr (in the mole ratio 2:2:1) and HNO3. The products (micro- and single-crystalline) were characterized by X-ray structure determination, FT-IR spectroscopy and TG/DSC analysis. The most striking structural feature of (Hdipya)(H3pyr) are short inter- and extremely short intramolecular hydrogen bonds. These bonds mutually connect cations and anions making thin layers parallel to the crystallographic (223) plane. In addition, quite unusual, linear proton-bound polymeric {H(H2pyr) -}n were identified in the structure. The results of crystal structure determination are compared with FT-IR and TG/DSC data. The low position of the nuas(COO) vibration at 1660 cm-1 is in accordance with strong hydrogen bonding. This value could be used as a measure of the C=O bond order, which was calculated to be 1.81.

Design, synthesis, and biological characterization of a caspase 3/7 selective isatin labeled with 2-[18f]fluoroethylazide

Imaging of programmed cell death (apoptosis) is important in the assessment of therapeutic response in oncology and for diagnosis in cardiac and neurodegenerative disorders. The executioner caspases 3 and 7 ultimately effect cellular death, thus providing selective molecular targets for in vivo quantification of apoptosis. To realize this potential, we aimed to develop 18F-labeled isatin sulfonamides with high metabolic stability and moderate lipophilicity while retaining selectivity and affinity for caspase 3/7. A small library of isatins modified with fluorinated aromatic groups and heterocycles was synthesized. A lead compound incorporating 2?- fluoroethyl-1,2,3-triazole was identified with subnanomolar affinity for caspase 3. “Click labeling” provided the 18F-labeled tracer in 65 ¡À 6% decay-corrected radiochemical yield from 2-[18F] fluoroethylazide. The compound showed high stability in vivo with rapid uptake and elimination in healthy tissues and tumor. The novel 18F-labeled isatin is a candidate radiotracer for further preclinical evaluation for imaging of apoptosis.
