N-Acetylneuraminate lyase and N-acyl-D-glucosamine 2-epimerase had been cloned and over-expressed in Escherichia coli. Simultaneous use of these two enzymes and feeding of appropriate amounts of pyruvate to the reaction mixture made possible the high conversion of N-acetylneuraminic acid (Neu5Ac) from N-acetyl-D-glucosamine (GlcNAc) with a 77% conversion rate on a molar basis. As a result, 29 kg of Neu5Ac was obtained from 27 kg of GlcNAc. The product was recovered by direct crystallization, and verified as identical to authentic Neu5Ac.

Design, synthesis, and antitubercular activity of 3-amidophenols with 5-heteroatomic substitutions

A series of novel 3-amidophenols with 5-heteroatomic substitutions were designed and synthesized. Several compounds showed potent antitubercular activity against Mycobacterium tuberculosis H37Ra (MIC = 0.25?5 mug/mL). Compounds 12j and 14i also displayed good inhibitory activity against M. tuberculosis H37Rv and two clinically isolated multidrug-resistant M. tuberculosis strains (MIC = 0.39?3.12 mug/mL). The privileged compound 14i showed certain oral efficacy on a mouse infection model. The compounds are non-cytotoxic against L-O2 hepatocytes and RAW264.7 macrophagocytes. They did not exert inhibitory activity against representative Gram-positive and Gram-negative bacteria.

Complex 5 can also be prepared by reaction of trans-Mo2(O2CCR3)2X 2(PPh3)2 with excess Hdpa. Their UV-Vis and IR spectra have been recorded and the structures of 4, 5 and 6 have been determined. The Hdpa ligand in 4 is coordinated to the metal centers via two terminal pyridine nitrogen atoms with a cis-N conformation. The dpa- ligands in 5 are bridged to the metal centers via one terminal nitrogen atom and the central nitrogen atom. The aligned-N conformation of the dpa- ligand is stabilized by both the intramolecular MoN and C-HN interactions.
