A novel three-enzyme reaction cycle for the synthesis of N-acetyllactosamine with in situ regeneration of uridine 5′-diphosphate glucose and uridine 5′-diphosphate galactose

A new three-enzyme reaction cycle consisting of sucrose synthase, UDP glucose 4′-epimerase, and human beta-1,4-galactosyltransferase was established for the synthesis of N-acetyllactosamine (LacNAc) with in situ regeneration of UDP galactose. We found that UDP glucose 4′-epimerase is reductively inactivated in the presence of UMP and acceptor substrates of beta-1,4-galactosyltransferase. Reactivation of UDP glucose 4′-epimerase by the transition state analogues dUDP or dTDP 6-deoxy-D-xylo-4-hexulose in combination with the repetitive batch technique enabled us to use the native enzymes for 11 days in this cycle. With 10 U of sucrose synthase, 5 U of UDP glucose 4′-epimerase, and 1.25 U of beta-1,4-galactosyltransferase, 594 mg of LacNAc could be synthesized. N-Acetyllactosamine was also subsequently converted to Neu5Acalpha2,6Ga1beta1,4GlcNAc with alpha-2,6-sialyltransferase and CMP-Neu5Ac.

All compounds were obtained as microcrystalline solids, stable in air and insoluble in common solvents. Two of them, [M(tpht)(dipya)(H2O)2]¡¤3H2O [M = Co(II), Ni(II); dipya = 2,2′-dipyridylamine], were obtained as single crystals and their crystal structures have been determined by X-ray structure analysis. Both structures consist of discrete [M(tpht)(dipya)(H2O)2]2+ complex units with two H2O molecules in the trans position. Three additional H2O molecules together with coordinated H2O and O atoms from COO groups make a three-dimensional network of hydrogen bonds. Terephthalato ligands are coordinated by only one COO group, which acts as a chelating ligand. Central metal ions are in a distorted octahedral environment. Based on the above-mentioned analysis and known crystal structures, the possible structures and geometry of the complexes are discussed. All Co(II) and Ni(II) complexes are pseudooctahedral and mononuclear, with only one of the tpht COO groups coordinated in a chelate mode. The structural characteristics of the ligands as well as the metal centers present in the neutral metallacycles allow easy tuning of their emission properties. Therefore, by engineering the different parts in neutral metal-organic ensembles, emissions from blue to red have been achieved. The neutral metallacyclic structures are unique in their well-defined forms and would be expected to function as a macrocycle host with a large internal cavity shapes generated by the tailored ligands. Hence, the presence of both photophysical properties and the cavity sizes of these neutral metallacycles make them intriguing hosts for neutral guests and metal ions. This review provides an overview of the structural features and properties of neutral discrete metal-organic cyclic architectures ranging from binuclear to fascinating multinuclear assemblies that emerged during the period from 2005 to 2012.

Selective Production of Terminally Unsaturated Methyl Esters from Lactones Over Metal Oxide Catalysts

Metal oxide catalysts were studied for their selectivity for the production of a terminally unsaturated methyl ester, methyl 5-hexenoate (M5H), from a 6 carbon, 6-membered ring lactone, delta-hexalactone (DHL). A 15?wt% Cs/SiO2 catalyst had a selectivity of 55% to M5H. This selectivity was the highest of the metal oxide catalysts studied, which were Cs/SiO2, MgO, SrO, CeO2, ZrO2, Ta2O5, MgAl2O4, and a Mg?Zr mixed oxide. The Cs/SiO2 catalyst was utilized for the ring-opening of gamma-valerolactone (GVL), a 5 carbon, 5-membered ring lactone. The catalyst was 88% selective to the terminally unsaturated methyl ester, methyl 4-pentenoate (M4P). Weight hourly space velocity studies determined that the unsaturated ester distributions remained constant and no C=C double bond isomerization occurred. Liquid phase transesterification reactions with DHL and methanol and nuclear magnetic resonance spectroscopy confirmed that DHL undergoes ring-opening transesterification to produce an?omega-1 hydroxy methyl ester, methyl 5-hydroxyhexanoate (M5HH). Liquid phase transesterification reactions and thermochemistry calculations established that the equilibrium for GVL transesterification with methanol was favored towards the ring-closed lactone instead of the ring-opened hydroxy ester because of the decreased ring strain of GVL compared to DHL. The difference in terminally unsaturated methyl ester selectivity between GVL and DHL manifests from the difference in ring-strain energy. DHL passes through the M5HH intermediate as a result of greater ring strain, while the production of M4P from GVL most likely occurs through a direct, concerted mechanism.
