Rational design of novel, potent small molecule pan-selectin antagonists

This report describes the first results of a rational hit-finding strategy to design novel small molecule antiinflammatory drugs targeting selectins, a family of three cellular adhesion molecules. Based on recent progress in understanding of molecular interaction between selectins and their natural ligands as well as progress in clinical development of synthetic antagonists like 1 (bimosiamose, TBC1269), this study was initiated to discover small molecule selectin antagonists with improved pharmacological properties. Considering 1 as template structure, a ligand-based approach followed by focused chemical synthesis has been applied to yield novel synthetic small molecules (MWr < 500) with a trihydroxybenzene motif, bearing neither peptidic nor glycosidic components, with nanomolar in vitro activity. Biological evaluation involves two kinds of in vitro assays, a static molecular binding assay, and a dynamic HL-60 cell attachment assay. As compared to controls, the novel compounds showed improved biological in vitro activity both under static and dynamic conditions. In this regard, P-Selectin glycoprotein-1 (PSGL-1) has been identified as a ligand for P-Selectin. PSGL-1 binds to P-Selectin through the interaction of core-2 O-glycan expressing sialyl Lewisx oligosaccharide and the three tyrosine sulfate residues. Herein, we report the synthesis of threonine-linked core-2 O-glycan as an amino acid building block for the synthesis of PSGL-1. This building block was further incorporated in the Fmoc-assisted solid-phase peptide synthesis to provide a portion of the PSGL-1 glycopeptide.

Design, synthesis and anti-influenza A virus activity of novel 2,4-disubstituted quinazoline derivatives

Four 2,4-disubstituted quinazoline series containing various amide moieties were designed and synthesized as new anti-influenza A virus agents using the strategies of bio-isosterism and scaffold hopping. Many of them exhibit potent in vitro anti-influenza A virus activity and low cytotoxicity (CC50: >100 muM). Particularly, compounds 10a5 and 17a show better activity (IC50: 3.70?4.19 muM) and higher selective index (SI: >27.03, >23.87, respectively) against influenza A/WSN/33 virus (H1N1), opening a new direction for quinazoline derivatives in anti-influenza A virus field.

The reactions and coordination geometry of bis(2-pyridyl)amine (Hdpa) and its deprotonated anion (dpa) can be effectively modified by the reaction conditions. Depending on the solvent system, the reaction of multinuclear ruthenium carbonyls such as Ru3(CO)2 and [Ru(CO) 3Cl2]2 with Hdpa at moderate reaction temperatures lead to low yields of a monomeric cis(CO), cis(Cl)-Ru(Hdpa)(CO)2Cl2 or a dimeric metal-metal-bonded [Ru(dpa)2(CO)2]2. In organic solvents high temperatures favor the formation of [Ru(dpa)2(CO)2] 2 from clustered Ru3(CO)12 and Hdpa. The high-temperature reactions in HCl solution can, in turn, be used for selective synthesis of cis(CO),cis(Cl)-Ru(Hdpa)(CO)2Cl2. dpa (deprotonated with CH3Li) readily reacts with [Ru(CO) 3Cl2]2, Ru3(CO)12, or H4Ru4(CO)12 in organic solvents, leading to [Ru(dpa)2(CO)2]2. Under the corresponding conditions Os carbonyls produce a new type of dpa-bridged dimer compound without a direct metal-metal bond, [Os(dpa)(CO)2(CH 3)]2, in addition to ruthenium dimer equivalent [Os(dpa)2(CO)2]2.

An ammonium sulfate sensitive chitinase from Streptomyces sp. CS501

A chitinase from Streptomyces sp. CS501 was isolated from the Korean soil sample, purified by single-step chromatography, and biochemically characterized. The extracellular chitinase (Ch501) was purified to 4.60 fold with yield of 28.74 % using Sepharose Cl-6B column. The molecular mass of Ch501 was approximately 43 kDa as estimated by SDS-PAGE and zymography. The enzyme (Ch501) was found to be stable over a broad pH range (5.0-10.0) and temperature (up to 50 C), and have an optimum temperature of 60 C. N-terminal sequence of Ch501 was AAYDDAAAAA. Intriguingly, Ch501 was highly sensitive to ammonium sulfate but it’s completely suppressed activity was recovered after desalting out. TLC analysis of Ch501 showed the production of N-acetyl d-glucosamine (GlcNAc) and Diacetylchitobiose (GlcNAc)2, as a principal hydrolyzed product. Ch501 shows antifungal activity against Fusarium solani and Aspergillus brasiliensis, which can be used for the biological control of fungus. As has been simple in purification, stable in a broad range of pH, ability to produce oligosaccharides, and antifungal activity showed that Ch501 has potential applications in industries as for chitooligosaccharides production used as prebiotics and/or for the biological control of plant pathogens in agriculture.

In this study, we isolated three kinds of antigenic polysaccharide components (tentatively designed as AP-1 ~ 3) from cells of Leptospira biflexa serovar patoc strain Patoc I (L. biflexa patoc Patoc I) by the hot phenol-water procedure, followed by treatment with mild acid and column chromatography. Two of them (AP-1 and AP-2) were recovered from the phenol-soluble fraction whereas another (AP-3) was recovered from the aqueous fraction. All of them reacted toward an anti-L. biflexa serum and also cross-reacted in similar extents toward most of the other leptospiral antisera tested. Such immunoreactions were specifically inhibited by a beta-(1 ? 4)-linked mannobiose, but were not by any mono- and oligosaccharide tested. From their structural analyses including 1H and 13C NMR spectrometry, Smith degradation and methylation analysis, it was revealed that all of these antigenic polysaccharides had the same disaccharide unit ?3)-beta-D-Manp-(1 ? 4)-beta-D-Manp-(1? in their major polysaccharide parts, but they differed in the acyl substituents. Therefore it is most likely that such mannobiose unit is a candidate for the antigenic epitopes of L. biflexa polysaccharides. (C) 2000 Elsevier Science Ltd.
