A facile and highly stereoselective synthesis of 1-thiotrehalose, that is, alpha,alpha-S-linked trehalose, is described. Glycosylation of configurationally pure alpha-glucosyl thiol 5 with glucosyl trichloroacetimidate 6 or glucosyl thioimidate 9 followed by deprotection afforded 1-thiotrehalose in excellent alpha-stereoselectivity and high yield. A different synthetic route to the key building block, alpha-glucosyl thiol 5, was also investigated in this report.

All three molecules in the two polymorphs differ significantly in their conformations, especially with respect to the methyl carboxylate and trichloroacetimidoyl groups.

The NaH-NaI composite exhibits unique chemoselectivity for reduction of N,N-dimethylamides over ketones.
