CXCR2 has emerged as a therapeutic target for not only peripheral inflammatory diseases but also neurological abnormalities in the central nervous system (CNS). Herein, we describe the discovery of a novel 1-cyclopentenyl-3-phenylurea series as potent and CNS penetrant CXCR2 antagonists. Extensive SAR studies, wherein molecules’ property forecast index (PFI) was carefully optimized for overall balanced developability profiles, led to the discovery of the advanced lead compound 68 with a desirable PFI. Compound 68 demonstrated good in vitro pharmacology with excellent selectivity over CXCR1 and other chemokine receptors. Rat and dog pharmacokinetics (PK) revealed good oral bioavailability, high oral exposure, and desirable elimination half-life of the compound in both species. In addition, the compound demonstrated dose-dependent efficacy in the in vivo pharmacology neutrophil infiltration “air pouch” model in rodents after oral administration. Further, compound 68 is a CNS penetrant molecule with high unbound fraction in brain tissue.

Antagonists of peripheral type 1 cannabinoid receptors (CB1) may have utility in the treatment of obesity, liver disease, metabolic syndrome and dyslipidemias. We have targeted analogues of the purine inverse agonist otenabant (1) for this purpose. The non-tissue selective CB1 antagonist rimonabant (2) was approved as a weight-loss agent in Europe but produced centrally mediated adverse effects in some patients including dysphoria and suicidal ideation leading to its withdrawal. Efforts are now underway to produce compounds with limited brain exposure. While many structure-activity relationship (SAR) studies of 2 have been reported, along with peripheralized compounds, 1 remains relatively less studied. In this report, we pursued analogues of 1 in which the 4-aminopiperidine group was switched to piperazine group to enable a better understanding of SAR to eventually produce compounds with limited brain penetration. To access a binding pocket and modulate physical properties, the piperazine was functionalized with alkyl, heteroalkyl, aryl and heteroaryl groups using a variety of connectors, including amides, sulfonamides, carbamates and ureas. These studies resulted in compounds that are potent antagonists of hCB1 with high selectivity for hCB1 over hCB2. The SAR obtained led to the discovery of 65 (Ki = 4 nM, >1,000-fold selective for hCB1 over hCB2), an orally bioavailable aryl urea with reduced brain penetration, and provides direction for discovering peripherally restricted compounds with good in vitro and in vivo properties.

Virtually complete regioselective galactosylation of the diol acceptor p-methoxyphenyl 6-O-benzyl-2-deoxy-2-tetrachlorophthalimido-beta-D- glucopyranoside (8) with the donor phenyl 2,3,4-tri-O-acetyl-6-O-benzyl-1- thio-beta-D-galactopyranoside (11) gave the lactosamine derivative 14, which was fucosylated with the donor 15 to give the Le(x) trisaccharide glycoside 2 after deprotection. Regioselective sialylation of the partially protected Le(x) trisaccharide triol 24 with the sialyl donor 25 gave, after deprotection, the SLe(x) tetrasaccharide glycoside 1. The overall yields of 2 and 1 from the monosaccharide starting materials 8, 11, 15, and 25 were 56% and 29%, respectively. In contrast to the virtually complete regio- and stereoselective galactosylation of 8, fucosylation with the benzyl-protected donor 15 gave the corresponding 1?3- and 1?4-linked disaccharides in a ratio of 3.6:1 (highly stereo- but not regioselective glycosylation), whereas fucosylation with acetyl-protected donor 18 gave a 2.2:1 beta/alpha-mixture of 4- O-linked disaccharides (highly regio- but not stereoselective glycosylation).

Glycosidation of the myo-inositol derivatives 2 and 3 by the diazirine 1 yields 90percent of a diastereoisomer pair of beta-D-glycosides in a 1:1 ratio, i.e. 5/6 and 7/8, respectively (Scheme 1).The crystal structure of 3 shows a strong intramolecular H-bond, which persists in solution, as indicated by FT-IR and 1H-NMR spectra.Yields and diastereoselectivity are lower for the glycosidation of 24 by 1 (Scheme 3).The resulting 1,2- and 1,4-linked disaccharides 25-28 were isolated as their acetates 29-32.The previously determined crystal structure of 24 shows no intramolecular H-bonds.The yield of the glycosidation of 24, but not of 3, depends upon the concentration, indicating that activation of 24 by intermolecular H-bonds is required.Glycosidation of 2 and 3 with the trichloroacetimidate 14 gave mixtures of four (5, 6, 15, and 16), and six (7, 8, and 17-20) disaccharides, respectively (Scheme 2).

Some cyanide-bridged complexes are known for exhibiting slow magnetic relaxation behavior in a light-induced metastable state. Herein, an unexpected reverse effect is observed for the first time in the S= 1/2 {FeIILS-CoIIILS-FeIIILS} (HS=high spin, LS=low spin) ground state of a novel V-shaped trinuclear cyanide-bridged {Fe2Co} complex. In this complex, light-switchable iron-cobalt charge transfer with repeatable off/on switching of slow magnetic relaxation is discovered upon alternating laser irradiation at 785 and 560 nm. An important characteristic of the present compound is that the S= 1/2 ground state exhibits slow magnetic relaxation before irradiation, whereas this is accelerated after irradiation. This is different from the typical behavior, where the light-induced metastable state exhibits slow magnetic relaxation.

Methylenecyclopropanes are important synthetic intermediates that possess strain energies exceeding those of saturated cyclopropanes by >10 kcal/mol. This report describes a catalytic reductive methylenecyclopropanation reaction of simple olefins, utilizing 1,1-dichloroalkenes as vinylidene precursors. The reaction is promoted by a dinuclear Ni catalyst, which is proposed to access Ni2(vinylidenoid) intermediates via C – Cl oxidative addition.

The structure of Bodipy-DPy was confirmed by UV-vis spectroscopy, fluorometry, melting point analysis, 1H-NMR, 13C-NMR, elemental analysis and ESI-TOF-MS. Its photophysical properties was investigated by absorption, emission and excitation techniques. Based on chelation assisted fluorescence enhancement, a fluorometric method was developed for trace level detection, selectivity and concentration studies of chromium(iii) in EtOH-H2O. The fluorescent ligand behaved as a chemosensor for Cr(iii) cations and showed a strong red fluorescence upon coordination with Cr(iii), whereas almost no fluorescence was observed for the other metal cations studied. The Bodipy-DPy has absorption and emission maxima at 528 nm and 535 nm, respectively. The equilibrium binding constant of Bodipy-DPy with Cr(iii) was calculated as 1.17 × 106 from the Stern-Volmer equation. This journal is
