Hemagglutinating Activity of Extracellular Alkaline Metalloendopeptidases from Vibrio sp. NUF-BPP1

Alkaline metalloendopeptidase (metalloprotease) AP1 (48 kDa) from Vibrio sp. isolated from the intestine of a five-barred goatfish (Parupeneus trifasciatus) was reported in our previous paper to produce AP2 (36 kDa) by releasing a peptide fragment (molecular mass of about 12 kDa) from the C-terminal end of AP1 by autodigestion.1) AP1 strongly agglutinated fish (flounder, Paralichthys olivaceus) and rabbit erythrocytes, and weakly chicken erythrocytes. In contrast, AP2 had no significant hemagglutinating activity toward any erythrocytes tested, except for weak activity on flounder erythrocytes, suggesting that the C-terminal region of AP1 may be required for the strong hemagglutinating activity. The optimum temperature for the hemagglutinating activity of AP1 was found to be lower than that for the proteolytic activity. At acidic pHs (below pH 7.5), the hemagglutinating activity of AP1 decreased, and its pH profile resembled that of the proteolytic activity. The hemagglutinating activity of AP1 was not observed in the presence of o-phenanthroline or synthetic and proteinous substrates, but different kinds of saccharides and lipids had no effect. While the proteolytic activity of AP1 was not affected by CaCl2, the hemagglutinating activity of AP1 decreased with increases in CaCl2 concentrations. These results suggested that the hemagglutinating activity of these proteases (AP1 and AP2) was most likely caused by their proteolytic action on erythrocyte cell surfaces.

Open versus Closed Polyaromatic Nanocavity: Enhanced Host Abilities toward Large Dyes and Pigments

Host functions of polyaromatic nanocavities were revealed by using an M2L4 molecular cage and capsule. On the basis of the previously reported M2L4 capsule with a closed polyaromatic cavity, a new M2L4 cage (as a mixture of the isomers) was prepared by the quantitative assembly of two metal ions and four desymmetrized bispyridine ligands with a single polyaromatic panel. The obtained, open nanocavity of the cage exhibited enhanced binding abilities toward large dyes and pigments in water. For example, two molecules of coumarin dyes were bound in the nanocavity and showed strong whitish emission (up to PhiF=34 %). Furthermore, metallopigments, the sizes of which are larger than the inner cavities of the cage and capsule, were bound only in the open polyaromatic nanocavity of the cage to give water-soluble 1:1 host?guest complexes.

Effect of isomalt as novel binding agent on compressibility of poorly compactable paracetamol evaluated by factorial design

Tablets containing paracetamol were prepared by wet granulation. The model drug, paracetamol (API) and the diluent, milled isomalt (IM) were applied in different ratios where the granulation liquid was kept constant in each sample. The purpose of the study was to investigate the effect of wet granulation with milled isomalt on the tabletting properties of poorly compressible paracetamol. The results of tabletting process and the tablet characteristics were evaluated with 32 type face-centered full factorial design. The API:IM ratio (X1) and compression force (X2) were selected as independent variables. The dependent variables were as follows: the parameters of the tabletting process – lubrication coefficient (R-value), relative elasticity, friction work – and characteristics of the dosage form such as tablet hardness, friability, disintegration time, tensile strength. The results indicated that milled isomalt has an advantageous effect on both the process of compression and the tablet characteristics – due to its binder properties. Furthermore an optimal composition of the API and the diluent is to be found according to the compression process and the dosage form.
