To develop novel PPARgamma ligands, we synthesized thirteen 3-{4-(2-aminoethoxy)phenyl}propanoic acid derivatives, which are designed based on the structures of rosiglitazone and 15d-PGJ2. Among these compounds, compound 9 was found to be as potent as rosiglitazone in a binding assay and a preadipocyte differentiation test. Molecular modeling suggested that the nonyl group of 9 interacted with hydrophobic amino acid residues constructing the hydrophobic region of PPARgamma protein where the alkyl chain of 15d-PGJ2 is expected to be located.

Structures of triterpenoids from the leaves of Lansium domesticum

From the methanolic extract of the leaves of Lansium domesticum, three new onoceranoid-type triterpenoids, lansium acids X?XII and a new cycloartane-type triterpene, lansium acid XIII, were isolated. The chemical structures of the isolated new compounds were elucidated on the basis of chemical/physicochemical evidence. For new onoceranoid-type triterpenoids, the absolute configurations were established by comparison of experimental and predicted electronic circular dichroism (ECD) data. Complex 5 can also be prepared by reaction of trans-Mo2(O2CCR3)2X 2(PPh3)2 with excess Hdpa. Their UV-Vis and IR spectra have been recorded and the structures of 4, 5 and 6 have been determined. The Hdpa ligand in 4 is coordinated to the metal centers via two terminal pyridine nitrogen atoms with a cis-N conformation. The dpa- ligands in 5 are bridged to the metal centers via one terminal nitrogen atom and the central nitrogen atom. The aligned-N conformation of the dpa- ligand is stabilized by both the intramolecular MoN and C-HN interactions. Substitution of the naphthalene ring affords an isoform selective JNK3 inhibitor, 30, with approximately 10-fold selectivity over both JNK1 and JNK2. A naphthalene ring penetrates deep into the selectivity pocket accounting for the differentiation amongst the kinases. Interestingly, the gatekeeper Met146 sulfide interacts with the naphthalene ring in a sulfur-pi stacking interaction. Synthesis and characterization of monosaccharide derivatives and application of sugar-based prolinamides in asymmetric synthesis

For the first time, the beta-anomer of N-acetylglucosamine derivative methyl 2-acetamido-3,4,6-tri-O-benzyl-2-deoxy-beta-D-glucopyranoside (9b) was synthesized, isolated, and used in the synthesis of sugar-based primary amine 4b. Sugar-based primary amine 5a, an anomeric pair of monosaccharide derivatives 6a and 6b, and precursors to all these compounds were also synthesized, isolated, and well studied by standard spectroscopic methods, including 2D NMR spectroscopy experiments such as 1H-1H COSY and HSQC. Then sugar-based primary amines 4a, 5a, 6a, and 6b were converted into the corresponding prolinamide organocatalysts (i.e., 1a, 2a, 3a, and 3b) in high yields. The catalytic activity of these sugar-based prolinamide organocatalysts was demonstrated in asymmetric aldol reactions in various solvents and at different temperatures. The oraganocatalyst 3a was shown to be an efficient and powerful organocatalyst for the enantioselective aldol reaction of various cyclic or acyclic ketones with aromatic aldehydes in a mild and facile reaction, which gave the corresponding aldol adducts with high levels of diastereoselectivity (up to 99:1) and enantioselectivity (up to >99 %) in almost all cases.
