Multi-dimensional systems built from dichromate anions – Syntheses, crystal structures, and magnetic properties

Three novel compounds with bridging dichromate anions with the formulae [Fe(4,4?-bpy)2Cr2O7]n (1), [Co(4,4?-bpy)2Cr2O7]n (2), and [Ni(dpa)2Cr2O7]¡¤H2O (3) (4,4?-bpy = 4,4?-bipyridine, dpa = 2,2?-dipyridylamine) have been synthesized and characterized. The isostructural compounds 1 and 2 belong to the monoclinic space group C2/c and have been shown to be three-dimensional polymers in which the dichromate anions connect neighboring layers formed by [M(4,4?-bpy)2]2+ (M = Fe, Co) units. Compound 3 is a metallacrown type compound, crystallizing in the monoclinic space group P2 1/n. Two chromate anions link two NiII centers forming a dinuclear entity. The uncoordinated oxygen atom of the dichromate anion forms a hydrogen bond with the N-H unit of dpa resulting in a two-dimensional network. Measurements of magnetic susceptibilities indicate the presence of antiferromagnetic interactions in 1 and 2 whereas the exchange coupling between the NiII ions in 3 is ferromagnetic. A comparison of the effects of 3-hydroxy-3-methylglutaryl-coenzyme a (HMG-CoA) reductase inhibitors on the CYP3A4-dependent oxidation of mexazolam in vitro

HMG-CoA reductase inhibitors can be divided into two groups: those administered as the prodrug, i.e., the lactone form (e.g., simvastatin and lovastatin), and those administered in the active form, i.e., the acid form (e.g., pravastatin, fluvastatin, atorvastatin, and cerivastatin). In this study, the influence of the lactone and acid forms of various HMG-CoA reductase inhibitors on metabolism by CYP3A4, a major cytochrome P450 isoform in human liver, was investigated by determining the in vitro inhibition constant (Ki value) using an antianxiety agent, mexazolam, as a probe substrate. In human liver microsomes, all the lactone forms tested inhibited the oxidative metabolism of mexazolam more strongly than did the acid forms, which have lower partition coefficient (logD7.0) values. In addition, the degree of inhibition of mexazolam metabolism tended to increase with an increasing logD7.0 value of the HMG-CoA reductase inhibitors among the lactone and acid forms. In particular, pravastatin (acid form), which has the lowest logD7.0 value, failed to inhibit CYP3A4 activity. Taking account of the lipophilicity of the inhibitors, in conjunction with the CYP3A4-inhibitory activity, could be very useful in predicting drug interactions between substrates of CYP3A4 and HMG-CoA reductase inhibitors.

Three chitinase isozymes, HoChiA, HoChiB, and HoChiC, were purified from the stomach of the greenling, Hexagrammos otakii, by ammonium sulfate fractionation, followed by column chromatography on Chitopearl Basic BL-03 and CM-Toyopearl 650S. Substrate specificities of these chitinases were compared with those of another fish stomach chitinase from the common mackerel, Scomber japonicus (SjChi), as well as two from the tobacco hornworm, Manduca sexta (MsChi535 and MsChi386). The efficiency parameters, kcat/Km, toward glycolchitin for HoChiA and SjChi were larger than those for HoChiB and HoChiC. The relative activities of HoChiA and SjChi toward various forms of chitin were as follows: shrimp shell or crab shell -chitin > beta-chitin ? silkworm cuticle -chitin. On the other hand, the relative activities of HoChiB and HoChiC were beta-chitin ? silkworm -chitin > shrimp and crab -chitin. MsChi535 preferred silkworm -chitin to shrimp and crab -chitins, and no activity was observed toward beta-chitin. MsChi386, which lacked the C-terminal linker region and the chitin-binding domain, did not hydrolyze silkworm -chitin. These results demonstrate that fish and insect chitinases possess unique substrate specificities that are correlated with their physiological roles in the digestion of food or cuticle.

Base-catalysed, one-step mechanochemical conversion of chitin and shrimp shells into low molecular weight chitosan

A facile, solid-state mechanochemical method was developed for the synthesis of low molecular weight chitosan (LMWC) in one step from chitin and crude shrimp shell powders, in which chitin undergoes simultaneous deacetylation and depolymerisation in the presence of a base catalyst under mechanical milling. The method is advantageous over traditional multi-step methods, featuring enhanced efficiency and significantly reduced environmental impact. Compared to traditional approaches, the base usage is reduced to about 1/10, and the molecular weight of the obtained LMWC product is much more narrowly distributed, with a polydispersity value of only 1.1. The degree of deacetylation and the molecular weight can be adjusted by varying the ball milling parameters. The influences of different types of bases were investigated by solid-state NMR analysis and control experiments, pointing out the critical role of base in both depolymerisation and deacetylation. Base-catalysed mechanochemical transformation of chitin and shrimp shells provides a solvent-free way to effectively valorize shellfishery waste for valuable products.

Ion-pair RP-HPLC determination of sugars, amino sugars, and uronic acids after derivatization with p-aminobenzoic acid

A new, selective, and sensitive ion-pair RP-HPLC method for the simultaneous determination of three classes of natural organic compounds, i.e., carbohydrates, amino sugars, and uronic acids, in environmental samples is presented, p-Aminobenzoic acid is used for precolumn derivatization of the analytes, enabling fluorescence (lambdaex 313 nm, lambdaem 358 nm) or photometric detection (303 nm). The dependence of the derivatization yield on the reaction conditions is examined. Derivatives of lactose, galactose, glucose, mannose, xylose, arabinose, galacturonic acid, glucuronic acid, N-acetylglucosamine, and glycerinealdehyde were separated on a RP-C18 column with hydrophilic end capping within 35 min, applying TBAHSO4 as the ion-pair reagent. The concentration detection limits range between 20 and 30 mug L-1 ((1-2) ¡Á 10-7 M) for fluorescence detection and between 30 and 75 mug L-1 for UV detection. A good linearity is achieved in the concentration range from 50 mug L-1 to 100 mg L-1 (r2 >0.99). The described method has been applied for the determination of mono-/disaccharides, uronic acids, and amino sugars in soil solutions and in landfill leachates.
