Copper-Catalyzed Formation of alpha-Alkoxycycloalkenones from N-Tosylhydrazones

The combination of 20 mol % of copper iodide and lithium tert-butoxide triggers the formation of a broad range of substituted, functionalized alpha-alkoxy 2H-naphthalenones from readily available N-tosylhydrazones. The data suggests that this transformation occurs through cycloaddition of a copper carbenoid with an ester, followed by a Lewis acid-catalyzed [1,2] alkyl shift of the in situ generated alkoxyepoxide intermediate. The combination of 20 mol % of copper iodide and lithium tert-butoxide triggers the formation of a broad range of substituted, functionalized alpha-alkoxy 2H-naphthalenones from readily available N-tosylhydrazones. Each of the four independent mercury(II) centers in 2 adopts a distorted trigonal bipyramidal environment coordinated by two pyridine N atoms of two different anionic dpa ligands, two nitrile N atoms of two mu1,5 bridged dca units and the fifth position is occupied by the amide N of one dpa. Cooperative intermolecular N-H¡¤¡¤¡¤N and C-H¡¤¡¤¡¤N hydrogen bondings promote dimensionality in 1. The compounds display intraligand 1(pi-pi) fluorescence in DMF solutions at room temperature.

Synthesis and antimicrobial activity of 6-sulfo-6-deoxy-D-glucosamine and its derivatives

6-Sulfo-6-deoxy-D-glucosamine (GlcN6S), 6-sulfo-6-deoxy-D-glucosaminitol (ADGS) and their N-acetyl and methyl ester derivatives have been synthesized and tested as inhibitors of enzymes catalyzing reactions of the UDP-GlcNAc pathway in bacteria and yeasts. GlcN6S and ADGS at micromolar concentrations inhibited glucosamine-6-phosphate (GlcN6P) synthase of microbial origin. The former was also inhibitory towards fungal GlcN6P N-acetyl transferase, but at millimolar concentrations. Both compounds and their N-acetyl derivatives exhibited antimicrobial in vitro activity, with MICs in the 0.125?2.0 mg mL?1 range. Antibacterial but not antifungal activity of GlcN6S was potentiated by D-glucosamine and a synergistic antibacterial effect was observed for combination of ADGP and a dipeptide Nva-FMDP.

Synthesis, characterization, thermal and DNA-binding properties of new zinc complexes with 2-hydroxyphenones

The neutral mononuclear zinc complexes with 2-hydroxyphenones (ketoH) having the formula [Zn(keto)2(H2O)2] and [Zn(keto)2(enR)], where enR stands for a N,N’-donor heterocyclic ligand such as 2,2?-bipyridine (bipy), 1,10-phenanthroline (phen) or 2,2?-dipyridylamine (dpamH), have been synthesized and characterized by IR, UV and 1H NMR spectroscopies. The 2-hydroxyphenones are chelated to the metal ion through the phenolate and carbonyl oxygen atoms. The crystal structures of [bis(2-hydroxy-4-methoxy-benzophenone)(2,2?-bipyridine) zinc(II)] dimethanol solvate and [bis(2-hydroxy-benzophenone)(2,2?- bipyridine)zinc(II)] dimethanol solvate have been determined by X-ray crystallography. The thermal stability of the zinc complexes has been investigated by simultaneous TG/DTG-DTA technique. The ability of the complexes to bind to calf-thymus DNA (CT DNA) has been studied by UV-absorption and fluorescence emission spectroscopy as well as viscosity measurements. UV studies of the interaction of the complexes with DNA have shown that they can bind to CT DNA and the corresponding binding constants to DNA have been calculated and evaluated. The complexes most probably bind to CT DNA via intercalation as concluded by studying the viscosity of a DNA solution in the presence of the complexes. Competitive studies with ethidium bromide (EB) have shown that the reported complexes can displace the DNA-bound EB, suggesting strong competition with EB for the intercalation site.

Pseudomonas aeruginosa flagellin protein recruits the mammalian host sialidase enzyme neuraminidase- 1 (NEU1) to remove sialic acid residues from the extracellular domain of the mammalian cell-surface protein MUC l (MUC l -ED), thereby exposing a cryptic binding site on the MUC l -ED protein backbone for flagellin binding. NEU1 -driven MUC l -ED desialylation rapidly increases P. aeruginosa adhesion to the airway epithelium. MUCl -ED desialylation also increases MUC l -ED cleavage and shedding from the cell surface, where desialylated, shed MUCl -ED competitively blocks P. aeruginosa adhesion to cell-associated MUCl -ED. Presented herein are data showing that exogenously-administered, deglycosylated MUCl -ED peptides reduced adhesion of P. aeruginosa to airway epithelial cells. Also presented are data showing that administration of P. aeruginosa to mice in combination with deglycosylated MUC 1 -ED decreased P. aeruginosa recovered from the lungs at 48 hr and 72 hr post-infection. Such findings are extended to the methods of treatment and prevention of bacterial infections defined herein.
