Kaempferol 3-O-(3?,6?-di-O-E-p-coumaroyl)-beta-d- glucopyranoside (1), an optimal metabolite of Scots pine seedlings for protection of deep-lying tissue against damaging UV-B, represents a typical acylated flavonol 3-O-glycoside. This compound was synthesized for the first time via two approaches. The first approach, starting with kaempferol, featured formation of the flavonol 3-O-glycosidic linkage with a glycosyl bromide under conventional PTC conditions. In the second approach, 5,7,4?-tri-O-benzyl- kaempferol was readily prepared from 2?,4?,6?- trihydroxyacetophenone and p-hydroxybenzoic acid, which was coupled with a glucopyranosyl o-hexynylbenzoate under the catalysis of a gold(I) complex to provide the desired 3-O-glycoside in excellent yield. A variety of the glycosyl o-hexanylbenzoates equipped with the 2-O-benzoyl group were also proven to be highly efficient donors for construction of the flavonol 3-O-glycosidic linkages.

When these receptors are crosslinked with antibodies or multivalent ligands, they transmit either activating or inhibitory signals. Here we describe binding and inhibition experiments with recombinant extracellular ligand-binding domains of the rat NKR-P1A (activating) and NKR-P1B (inhibitory) receptors. We did not observe any difference in the binding of monosaccharide ligands by these receptors, but revealed dramatic differences in the binding of oligosaccharides and glycodendrimers. The preparation, crystal structures and magnetic properties of three different copper(II) chains of formula [Cu(pyim)-(H2O)(dca)]n(NO3), (1), [Cu(dpa)(dca)2]n (2) and [Cu(bpa)(dca)2], (3) [pyim = 2-(2-pyridyl)imidazole, dca = dicyanamide anion, dpa = 2,2?-dipyridylamine and bpa = 1,2-bis(4-pyridyl)ethane] are reported. A chain structure with single, symmetrical, end-to-end dca bridges is found in compound 1. This bridging mode, with two short, equatorially coordinated Cu-N bonds has not been previously observed in the [Cu(dca)Xn] (X = coligand) family of compounds. The copper atom in 1 has a distorted square pyramidal geometry with a bidentate pyim ligand and two nitrile nitrogen atoms from two dca groups in the equatorial positions and a water molecule occupying the axial site. In compound 2, the single dca bridge is of the asymmetric end-to-end type. The copper environment in 2 is also distorted square pyramidal with a bidentate dpa ligand and two nitrile nitrogen atoms (one from a terminally bound dca and the other from a bridging dca) in the equatorial positions and a nitrile nitrogen atom from another bridging dca occupying the axial site. Compound 3 also features a uniform chain structure. In this case double, asymmetric end-to-end dca bridges and a bis-monodenate bpa bridge in the gauche conformation connect the neighbouring copper atoms. The copper(II) coordination geometry in 3 is elongated octahedral with two trans coordinated bpa nitrogen atoms and two dca nitrile nitrogens in the equatorial sites and two nitrile nitrogen atoms from two other dca groups occupying the axial positions. The copper-copper separations across the dca bridges are 7.7011 (9) (1), 7.689(2) (2) and 7.095(3) A (3). Magnetic susceptibility measurements for 1-3 in the temperature range 1.9-290 K reveal the occurrence of weak intrachain antiferromagnetic interactions [J = -0.35 (1), -0.10 (2) and -0.22 cm-1 (3) the Hamiltonian being defined as H= -J ?iSi¡¤Si + 1]. Macoma birmanica agglutinin recognizes glycoside clusters of beta-GlcNAc/Glc and alpha-Man

Macoma birmanica agglutinin (MBA) that seems to play crucial roles in the innate immunity of marine bivalve, M. birmanica has been earlier defined as GlcNAc/Man specific. However, most complementary carbohydrate structures to its binding domain and ligand clustering in its recognition profile have not been established. In this study, the complete recognition profile of MBA was examined by enzyme-linked lectin-sorbent assay and inhibition assay. Among the monosaccharides tested, GlcNAc was more reactive followed by Man and Glc, others were non-reactive; revealing the importance of equatorial -NAc group at C-2, -OH group at C-4 and C-6, and pyranose conformation of hexose. Moreover, beta-glycosides of GlcNAc and Glc were more potent whereas for Man it was alpha-glycoside. MBA recognized both exposed and internal alpha-Man and beta-GlcNAc/Glc residues well with most linkages except (beta1-4). This binding pattern was further extended and confirmed by polyvalent glycoside clusters of GlcNAc(beta1-2)Man(alpha1-, which was a better inhibitor than Man(alpha1-2/3/6)Man(alpha1- or Man(alpha1-3/6)Man(beta1- present in well-defined naturally occurring glycoproteins. This broad range specificity explains the importance of MBA as an important pattern recognition molecule that provides more realistic picture of carbohydrate-based immune response triggering.

A convenient preparation of several N-linked glycoamino acid building blocks for efficient solid-phase synthesis of glycopeptides

A high yielding route for the preparation of several Boc- and Fmoc-protected N-linked glycopeptide monomers was presented. It was found that these building blocks can be used for the solid-phase synthesis of glycopeptides or glycopeptidomimetics. A number of short glycopeptides- collagen mimics was prepared to demonstrate this applicability. The protocol employed was expected to be suitable for the synthesis of any desired N-linked glycopeptide.
